Control method

How to operate this laboratory glass fermenter system:
1.Preparatory work before fermentation
1)Check whether the power supply is normal.
2) Check the system valve, connector and fastening screws whether are tight.
3) Check the sealing performance of the fermentation tank, breathing filter, pipeline and valve whether are in good condition, whether have leakage. In-tank whether is sealing (should focus on testing when seasons change) , to ensure that all valves are closed.
4) Check the voltage and air pressure whether can be supplied normally. Input power is AC220V ± 10%, 50Hz frequency.
5) PH electrode operating instructions, please check the relevant specifications.
6) Check the motor whether can run normally.
2.Sterilization
1)The first cleaning system after fermentation system finish installed:
Tank inner cleaning: could remove the flange which is at the top of the tank, operators could use clean cloth to do manual cleaning, after finish should discharge all the waster water out,the use water washing a few times more. If the fermenter long time don’t use or cultivate bacteria is not same with last time, could use 2% NaOH cleaning, after cleaning could do fermentation system sterilization.
2)Real tank sterilization:
After finish preparing the culture medium, put it in the tank, remove: motor line and motor, temperature probe line, the air intake pipe,etc, which are connected to the tank parts. Install PH electrode, ORP electrode and DO electrode (screw on protection functional nuts), put the fermentation tank in the high pressure sterilization pot sterilization thoroughly.
3.Fermentation
After natural cooling of the complete sterilization fermenter and the culture medium, install the motor, PH electrode cable, DO cable, ORP cable, temperature electrode. Sterile air ventilation (the pressure of glass tank shoule not be larger than 0.05Mpa, the vent valve could keep open), on the operating interface to set the related fermentation process control parameter values.
4.Process sampling
By peristaltic pump suction or charge a trace of air with pressure in tank, open the silicone tube clip.
5.Material discharging
After the fermentation process, shut off the gas source, power supply, material feeding, the fermentation finished.
6.Maintenance and repair
1)The Fermenter should be placed in a clean, dry, well ventilated environment, the water and steam shall not directly splashed ove on the electrical parts.
2) After you use the fermenter, must be timely cleaning, prevent fermented liquid stem node within the fermentation tank, pipeline and valve.
3) All kinds of instruments, should according to the requirement to maintain and storage, pressure gauge, safety valve, temperature instrument should be calibrated once a year.
4) When to stop using the equipment should be clean and dry. The filter element of filter should be clean, dry and properly kept.
5) The surrounding environment of air compressor should be kept clean, the air is fresh, the drain for the living water, the fermenter body surface clean.
7.Matters need attention
1)When the sterilizing filter sterilization, the pressure shall not exceed 0.13 MPa.
